Introduction
It is generally accepted that cholesterol excess constitutes a major risk factor for development of different osteopathic states. However there’s little data on the effects of cholesterol excessive diet on mineral ultrastructure of bone tissue.
Aim of the study
The study was aimed at experimental investigation of ultrastructure of cancellous bone mineral in rats on high-cholesterol diet.
Materials and methods
For the purpose of the experiment we selected 70 female rats with the initial mass of 330-360 grams aged over 24 moths. In the experimental group the standard diet was modified with addition of cholesterol (2,5%) and animal (lard) fat (10%) and corresponding amylum deficiency. The observation terms were the 7th, the 15th, the 30th, the 90th and the 180th days of the experiment.
To verify the model we measured total serum cholesterol. X-ray tests of the ground bone obtained the proximal epiphysis of tibia where cancellous bone prevails were performed using DRON-2.0 X-ray diffractometer with GUR-5 goniometer. For these tests we used voltage and amperage 30 kV and 20 A. From the whole data we calculated interplanar distance and the parameters of the elementary cell of bone mineral, the coherent scattering blocks and microtexture coefficient.
Results
The results obtained allow conclusion that cholesterol excess that results in hyperholesterolemia also has a negative effects on ultrastructure of bone mineral. The changes manifest as crystallite size increase and decrease of their orientation uniformity in the crystal lattice beginning from the 15th day of observation which suggest d third stage of bone matrix mineralization that is crystal growth stage.
From the 90th day the mineralization processes become deranged in the second stage, i. e. in the stage of formation of the crystal nuclei which is manifested as increased elementary cells sizes along a-axis and c/a ratio. Such changes should be considered as progressing interplanar consolidation and conditioning of further destabilization of hydroxyapatite elementary cells, which leads to increased bone senescence rate.
Conclusions
In the study we demonstrated that cholesterol excessive diet results in deranged mineralization processes in bones and accelerated bone system senescence.

Wstęp
Ogólnie przyjmuje się, że podwyższony cholesterol jest głównym czynnikiem ryzyka rozwoju różnych zmian osteopatycznych. Jednakże istnieje niewiele danych na temat wpływu diety wysokocholesterolowej na ultrastrukturę mineralną tkanki kostnej.
Cel badania
Celem pracy było eksperymentalne zbadanie ultrastruktury kości beleczkowej u szczurów na diecie wysokocholesterolowej.
Materiał i metody
W celu przeprowadzenia eksperymentu wybraliśmy 70 samic szczura o początkowej wadze 330-360 gramów w wieku powyżej 24 miesiące. W grupie eksperymentalnej standardową dietę zmodyfikowano dodatkiem cholesterolu (2.5%) oraz tłuszczu zwierzęcego – smalcu (10%) a także odpowiednim niedoborem skrobii. Obserwacji dokonano w terminach: 7, 15, 30, 90 i 180 dni od rozpoczęcia eksperymentu.
W celu weryfikacji modelu dokonywano pomiarów cholesterolu w surowicy. Za pomocą rentgenowskiego dyfrakcjometru DRON-2.0 oraz goniometru GUR-5 wykonano badania rtg bliższej nasady kości piszczelowej gdzie dominuje kość beleczkowa. Do badań rtg użyto napięcia 30 kV i natężenia 20A. Ze wszystkich danych obliczono odległość międzypłaszczyznową oraz parametry podstawowych komórek kostnych, a także koherentny rozkład bloków i współczynnik mikrotekstury.
Wyniki
Uzyskane wyniki pozwalają na wniosek, ze podwyższony cholesterol, którego skutkiem jest hipercholesterolemia ma również negatywny wpływ na ultrastrukturę mineralną kości. Zmiany manifestują się jako zwiększenie rozmiarów kryształów hydroksyapatytu oraz zmniejszenie zwartości ich orientacji w siatce krystalicznej poczynając od 15 dnia obserwacji, co sugeruje zaburzenie trzeciego stopnia mineralizacji macierzy kostnej, tj. stadium wzrostu kryształów.
Poczynając od 90 dnia obserwacji proces mineralizacji był zaburzony w drugim stadium, tj. w stadium formowania jąder komórkowych kryształów, co manifestowało się wzrostem rozmiarów podstawowych komórek wzdłuż osi a i współczynnika c/a. Takie zmiany powinny być uznane za postępującą konsolidację międzypłaszczyznową i uwarunkowanie dla dalszej destabilizacji podstawowych komórek hydroksyapatytu, co prowadzi do zwiększenia wskaźnika starzenia się kości.
Wnioski
W badaniu wykazano, że wskutek diety wysokocholesterolowej następują zaburzenia procesu mineralizacyjnego kości i przyspieszony zostaje proces starzenia się kości.
